Tsinghua University has successfully developed China's first intelligent multifunctional MBP fusion heparinase

Under the support of key projects of the National Natural Science Foundation of China, the Institute of Biology and Chemical Engineering of Tsinghua University successfully developed China's first intelligent multifunctional enzyme, MBP (maltose-binding protein) fusion heparinase. After six months of pilot tests, the project team has steadily prepared ultra low molecular weight heparin with an average molecular weight of 2000 to 3000 that has reached the international advanced level.
Enzymatic catalysis is a key technology for building green chemistry. For a long time, China's biochemical drug for preventing and treating thrombosis, low molecular weight heparin, relies heavily on imports, and prices remain high, so the study is of great social significance. According to reports, the research team used biochemical technology to establish a new process for producing highly-efficient, intelligent multi-functional MBP fusion heparinase. They first added the fluorescent protein signal to the enzyme and could quickly trace it. Secondly, the easily separated MBP protein was added to the enzyme. In this way, the heparinase obtained in the experiment can be separated into high purity by only one step.
After half a year of industrial production tests, the MBP fusion heparinase developed by the institute can not only effectively degrade heparin, but also can prepare ultra low molecular weight heparin with an average molecular weight of 2000-3000 by controlling the enzyme reaction time and the amount of enzyme. In particular, it is worth mentioning that the preparation of low molecular weight heparin by enzymatic reaction conditions is mild, compared with the traditional chemical lysis method, it has the advantages of high yield, environmental friendliness, and easy to achieve continuous production. In addition, the obtained product and the heparinase are easily separated, and the enzyme can be used repeatedly, thereby reducing the cost of the enzyme and the production cost of the low molecular weight heparin. This intelligent multifunctional MBP fusion heparinase has applied for 3 patents, and won the silver prize at the 17th National Invention Exhibition in November 2007.
It is understood that this group is currently using this intelligent multifunctional enzyme for the transformation of phosphotransferases and industrial enzymes. In the case of phosphotransferases used in the fine chemical industry of food additives, the food additives produced by domestic chemical methods are toxic, while the biological methods monopolized by Japan and Korea are tracked by intelligent multifunctional phosphotransferases and can be monitored at any time. The activity of the enzyme, and the reaction process is mild, clean and pollution-free.
